I have a jailbroken iPhone 3GS running iOS 5.0.1 and would like to update it to the latest firmware that can be jailbroken. Will I need to re-jailbreak the device after the update is complete?

Yes, you will need to re-jailbreak your iPhone 3GS after updating the firmware. When you update iOS, the system files are overwritten, which removes the jailbreak. The final firmware version supported by the iPhone 3GS is iOS 6.1.6. You can update to this version via iTunes and then use a utility like p0sixspwn to apply a new jailbreak. Keep in mind that if you are using a software unlock like ultrasn0w to use the phone on different carriers, it may not be compatible with iOS 6.1.6. In that case, a factory IMEI unlock is the better long-term solution.
