I am looking for information on how to jailbreak and unlock an iPhone 3GS running iOS 6.0.1 (build 10A523). My device is a newer model (MC640LL) with the 05.16.07 baseband. Some sources say I cannot upgrade the baseband to 06.15.00, while others say it is possible. Please provide specific guidance for this hardware revision.

For an iPhone 3GS running iOS 6.0.1, you should update to iOS 6.1.6, as it is the final firmware version for your device. You can perform an untethered jailbreak on iOS 6.1.6 using the p0sixspwn tool. Regarding the unlock, your baseband version 05.16.07 does not support a software unlock via Ultrasn0w. You must not attempt to flash the 06.15.00 iPad baseband, as your serial number (starting with 143) indicates a manufacture date in late 2011. Flashing that baseband on units made after week 34 of 2011 will permanently brick the cellular radio. The only safe way to unlock your specific model is through an official IMEI unlock.
